イエローレーザー搭載機種: MACSQuant® VYB
MACSQuant VYBはMACSQuant Analyzer 10の便利な自動機能はそのままに、イエローレーザーを搭載したフローサイトメーターです。近年さまざまな蛍光タンパク質が開発され、マルチカラー解析のオプションが広がっています。MACSQuant VYBはそれら新規色素に対応したフローサイトメーターです。
561 nmレーザーを搭載し、8蛍光色素を検出可能なMACSQuant VYB はmCherry, DsRed, GFP, CFPなどさまざまなフルーツ蛍光タンパク質標識の抗体や、その他の通常の蛍光色素標識の抗体の検出に理想的なフローサイトメーターです。さらに、PEの検出感度を高めるために、PEはイエローレーザーで励起し、FITCはブルーレーザーで励起する光学システムを採用しました。

3レーザー(405, 488, 561 nm)および、2つのスキャッターチャンネル (FSC, SSC) と8つの蛍光チャンネルを搭載したMACSQuant Analyzer VYB は全自動マルチカラー解析が可能な理想のフローサイトメーターです。
MACS MiniSamplerと組み合わせれば、最大96サンプルの処理を自動で行うことが可能です。MACS MiniSamplerは各種遠沈管やマイクロタイタープレートに対応し、さらに自動サンプル染色を行う場合は、4種類までの試薬バイアルも載せることができます。
MACSQuant ColumnはMACSQuant Analyzer VYBに内蔵されており、希少細胞の解析を迅速かつ効率的に行うために、解析前にサンプルを濃縮することが可能です。数ステップで簡単に濃縮できます。
弊社の抗体および各種キットとMACSQuant Analyzer VYBと組み合わせれば、樹状細胞、内皮前駆細胞、ガン幹細胞、サイトカイン分泌細胞、抗原特異的T細胞、末しょう循環腫瘍細胞などをより効率的に細胞解析が可能です。
細胞の絶対数測定は内蔵されたシリンジポンプにより正確にサンプル体積を測定・採取して行われます。結果は「events/μL」の形に自動計算されて表示されます。測定は1分以内に終了し、高価な内部標準ビーズは不要です。

The MACSQuant VYB can detect a wide range of fluorescent proteins. Histograms of CHO cells transfected with CFP, GFP, YFP (top row), mCherry, and mKate2 (bottom row) are easily distinguished from non‑transfected cells (shown in purple). GFP and YFP are often difficult to distinguish using flow cytometry due to similarities in their emission spectra. With the MACSQuant VYB, YFP‑ (shown in yellow) and GFP‑expressing cells (shown in green) are clearly distinguishable when observed in the PI channel (bottom right), making it possible to detect up to five fluorescent proteins in a single sample.

Excitation of both PE and GFP with the 488 nm blue laser only minimally resolves the two signals (A).
