Overview この抗体は、フローサイトメトリーまたは蛍光顕微鏡によるCD107a+ 細胞の同定と解析に使用できます。また、ヒトサイトカインに対する抗体と組み合わせて、刺激したCD8+ T細胞の細胞内サイトカイン発現の解析にも使用できます。 Details Background information クローンH4A3は、110-140 kDa のI 型膜糖タンパク質でlysosome-associated membrane protein 1 (LAMP-1)としても知られる、CD107a抗原を認識します。これは広く存在する細胞内タンパク質で、リソソームやエンドソームの膜に存在します。一過性に細胞膜上に存在するCD107aは、刺激によるCD8+T細胞の脱顆粒のマーカーとして使用できます1。また、CD8+T細胞よりも程度は低いものの、活性化NK細胞にも発現しています。 Clone Isotype H4A3 Mouse IgG1		Human PBMCs were stimulated in vitro for 5 hours with SEB in the presence of monensin and CD107a antibodies conjugated to VioBlue (A), FITC (B), PE (C), or APC (D). Cells were harvested, fixed, permeabilized, and intracellularly stained with Anti-IFN-γ antibodies. Additionally, cell surface staining was performed with CD8 antibodies. Cells were analyzed by flow cytometry using the MACSQuant® Analyzer. CD8+, IFN‑γ–secreting cells are shown. A B C D
