NK Cell Activation/Expansion Kit, human
Description NK Cell Activation/Expansion Kit は、ヒト NK細胞を活性化し、増殖させるために開発されました。このキットには、Anti-Biotin MACSiBead™ Particles と、ヒト CD335 (NKp46) および CD2 に対するビオチン化抗体が含まれています。ビオチン化抗体をロードした Anti-Biotin MACSiBead Particles は、ヒト血液もしくは PBMCs から分離した休止期NK細胞を、活性化し増殖させるために使用します。 Applications Anti-Biotin MACSiBead Particles を用いて増やした NK細胞は、サイトカインの解析や細胞傷害活性、遺伝子発現、機能解析などのアプリケーションに用いることが出来ます。		Figure 1 Anti-Biotin MACSiBead Particles were loaded with CD335 (NKp46) and CD2 antibodies. NK cells were isolated using the NK Cell Isolation Kit, human, and expanded using one loaded Anti-Biotin MACSiBead Particle per two NK cells. NK cells were cultured in medium supplemented with 10% FCS and 500 IU/mL rIL-2 at an initial density of 106 NK cells per mL. Cells were expanded for 3 weeks. For comparison, NK cells were cultured in medium supplemented with 10% FCS and 500 IU/mL rIL-2 alone. Figure 2 Purified human NK cells were activated and expanded using the NK Cell Activation/Expansion Kit. After 2 weeks of culture in medium supplemented with 10% FCS and 500 IU/mL rIL-2, cytotoxicity of NK cells was determined by incubation with K562 target cells in different ratios. In addition, cells incubated in medium supplemented with 10% FCS and 500 IU/mL rIL-2 alone and freshly isolated NK cells were analyzed. Specific target cell lysis by NK cells was determined after 4 hours.
